Ambientato alla fine del 18° secolo, durante la guerra di indipendenza americana, Assassin's Creed III introduce un nuovo eroe, Ratohnhaké:ton, di origini sia native americane che inglesi. Dopo aver adottato il nome di Connor, il protagonista diventa la nuova voce della giustizia nell'antico conflitto tra Assassini e Templari. I giocatori assumeranno così il ruolo di un Assassino nella guerra per la libertà contro la spietata tirannia degli inglesi e vivranno una delle esperienze di combattimento più realistiche e intense mai apparse nella serie. 

La trama di Assassin's Creed III si svolge durante la Rivoluzione Americana e condurrà i giocatori da remote terre di confine a vivaci città coloniali, fino a caotici campi di battaglia, dove l'Esercito Continentale di George Washington affronterà le imponenti truppe inglesi.
